I have a M100 with v1.3.1.00 firmware, single controller and X3 camera.  The X3 camera is from my Inspire 1 so I know the camera works.

I have R/C control of the M100, however, the camera control switches/buttons on the DJI Go app ar greyed out, there is no video signal, and the gimbal does not power on when the aircraft is powered on.

I know the camera works because I upgraded the firmware on the M100 using the X3 camera, and during that process the gimbal moved the camera, however, since upgrading the firmware, the gimbal does not power on when the system is powered on.

I have tried to downgrade the firmware to 1.2.80 using the DJI Assistant app on my PC, and during the upgrade the gimbal becomes powered on, but after the firmware downgrade is completed, the gimbal doesn't power on again.  I have then re-upgraded back to 1.3.1.00

Just to clarify, were you moving the gimbal via the RC or did you mean that it was just initializing as normal? The X3 still works fine when connected to your Inspire, correct? When you upgraded firmware via the X3, did you happen to keep the LOG file from that update. If so, could you provide that? Do you use any onboard SDK with this unit, or just the Go app?

In APP, Under HD make sure that EXT is on and not LB.

Yes, the X3 cameras (I have 2 of them), work fine with Inspire 1 and Inspire 1 pro.  The M100 is just a plain M100 with X3 camera, and DJI Go app

I'll check this setting.  I'm not aware of it, I had assumed that M100 + X3 camera was pretty much going to be like Inspire1.  I'll check when I get home.

The X3 camera doesn't initialise (the gimbal doesn't seem powered) when I power on the drone.  The only time the gimbal gets powered up is when I am using DJI Assistant to upgrade/downgrade the firmware.

I don't see EXT or LB options in the Go App.  Just Auto/Custom 60/50HZ, Simultaneous APP/HDMI, and OSD on HDMI as options in the HD tab.

I'm just using the Go App with the M100 + X3 camera (tried two different X3 cameras now) and tried reinstalling the Go app.  The gimbal just doesn't power on when I turn on the M100, and there is no video, and the camera controls are greyed out.  The Video Signal is showing in the Go app as 5/5 bars

Just to make sure I'm on the same page, was the M100/X3 set up working before or is this during the installation? If it's the former, did you recall this happening after a firmware update or a hard landing?

The aircraft has never been flown.  The drone firmware was update by using microsd card in the camera.  Once complete, the camera and gimbal never powered again on the drone, except at the 67% mark when the drone was downgrade and also upgraded using DJI Assistant.

After you performed the firmware update via the SD card, did you reformat the SD card? Just to make sure I'm  on the same page, you could update the aircraft via the Assistant 2 software, but the issue persists?

The only thing that has fixed this problem is replacing the Gimbal adaptor kit with a new one.

I would agree that this would be the next step.
