Hello,

While flying the P4, how should we position the antennas (with or without the signal boosters) to ensure the maximum signal strength?
How far we should stand?

This is explained in the manual.   Parallel to each other is best.   once everything is warmed up it doesn't matter where you stand.  
If you can go online,  you can find many videos that will show you.
There are tutorials on this forum menu that give instructions on many  aspects of DJII products.
