Dji Osmo Mobile Works fine With Samsung Galaxy S6. But - in front of the handle, there is a Jack-hole for microphone. I can't use any external microphone! How get this to work?

That's because it is not microphone connector. The 3.5mm jack is charging connector. ;)

For DJI devices, reading the manual is sooo important.  Lots of things in these devices are not what they might appear to be.  Also, user tips are critical with them.

It is not a mic input.
Check the manual
Just use the S6 mini jack if it fits.
Otherwise use a bluetooth mic.
